目的:探讨表皮生长因子受体(EGFR)基因第一内含子区rs763317位点单核苷酸多态性( SNP) 与食管癌放射敏感性的相关性.方法:随机选取160例食管癌患者,采用MassARRAY SNP分型技术分别检测其外周血EGFR基因多态位点rs763317的基因型,再用PCR-RFLP法检测这些基因型的SNP,进一步统计分析EGFR的rs763317位点SNP在食管癌放射治疗有效组、无效组中的分布情况.结果:160例食管鳞癌患者放疗的有效率为50%,AA等位基因在食管癌放射治疗有效率为65.5%,与无效组之间的分布差异具有显著的统计学意义( P < 0.001),而GA、GG等位基因在两组间分布的差异均无统计学意义(P>0.05).结论:AA的SNP与食管癌放射敏感性存在相关性,而GA、GG无相关性.%Objective: To investigate the association between single nucleotide polymorphism ( SNP ) rs763317 in genetic variations of the epidermal growth factor receptor ( EGFR ) and the clinical sensitivity in patients with esophageal cancer treated with radiotherapy. Method: A total of 160 esophageal cancer patients treated with radiotherapy were analyzed. Polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP ) and MassARRAY SNP methods were conducted to analyzed the genotypes. Results: The effective rate of radiotherapy 160 patients was 50% . Rs763317 was associated with the efficacy of radiotherapy . The response rate for the rs763317 AA was significantly higher than that for the GA or GG ( 65. 5% vs 9. 1% , P<0. 001 ). Conclusion: rs763317 polymorphism is associated with the clinical efficacy of radiotherapy and may be a potential biomarker to predict the clinical outcome in esophageal cancer patients treated with radiotherapy. There's no correlatron between GA, GG and sensitivity of radiotherapy in patients with esophageal cancer.
