红蓝草紫色素分离纯化研究

         

摘要

以红蓝草(Peristrophe baphica Bremk)茎叶为原料,用溶剂萃取法提取紫色素,用大孔树脂吸附法分离紫色素,进而探讨红蓝草紫色素的吸附与分离条件。结果表明,根据吸附前后溶液吸光度和颜色的变化,选择X-5、D4020、D101、S-8作为红蓝草紫色素的吸附树脂;洗脱溶剂以乙醇为佳,且乙醇体积分数以75%为宜;乙醇洗脱效果为D101﹥X-5﹥D4020,而S-8吸附的色素很难被乙醇洗脱。进行色素柱层析分离时,X-5和D101大孔树脂分离富集红蓝草紫色素的效果优于D4020大孔树脂。%Abstrct: The purple pigments of Peristrophe baphica Bremk stem and leaf, were extracted by solvent extraction method and purified by macroporous resin, and the adsorption and separation conditions were discussed. The results showed that X-5, D4020, D101, S-8 as adsorption resin of P. baphica Bremk purple pigment according to the before and after the adsorption of absorbance and color changes; and 75% ethanol should be used for desorption solvent and the desorption effect was D101﹥X-5﹥D4020, while the purple pigments adsorbed by S-8 were difficult to be desorbed; in addition, when enriched by col-umn chromatography, the enrichment effect for X-5 and D101 were better than D4020. To sum up, X-5 for adsorption and 75% ethanol for desorption were the optimal condition for separation of purple pigment from P. baphica Bremk.
