高效液相色谱法快速检测血管紧张素转换酶抑制活性

摘要

建立高效液相色谱法快速检测血管紧张素转换酶(ACE)抑制活性的方法。色谱条件为：以ODS C18柱(150mm×4.6mm，5μm)为色谱柱，乙睛-水(20：80，V/V，含0.5‰甲酸)为流动相，流速1mL/min，检测波长为228nm。结果表明：马尿酸在0.1～500μg/mL质量浓度范围内线性关系良好，相关系数r=0.9999；平均回收率为89.77%～99.57%，RSD为0.01%～1.52%。不同的ACE抑制剂检测结果证明该方法操作简便快速、重现性好、准确度高，可以作为ACE抑制剂体外活性的检测方法。%A rapid HPLC method was developed for the determination of angiotensinⅠ-converting enzyme (ACE) inhibitory activity. Hippuric acid was separated on an ODS C18 column (150 mm × 4.6 mm, 5μm) using a mobile phase consisting of acetonitrile and water (20:80, V/V, containing 0.5‰methanoic acid) at a lfow rate of 1 mL/min and detected at 228 nm. A good linear relationship was observed for hippuric acid over the concentration range of 0.1 to 500μg/mL with a correlation coefifcient of 0.9999. The average spike recovery rates of hippuric acid were 89.77%–99.57%with a relative standard deviation (RSD) of 0.01%–1.52%(n=3). The HPLC method was simple, rapid, reproducible, accurate and suitable for the analysis of ACE inhibitory activity in vitro as demonstrated by its applications to determine different ACE inhibitors.
