    • 摘要: Klinefelter syndrome represents one of the most common male sex chromosome disorders with a prevalence of 1 out of 500 male newborns[1].The syndrome is marked by the presence of an additional X chromosome in human males.It manifests mostly as 47,XXY,and can take the mosaic form as 46,XY/47,XXY.A 36-year-old adult had been referred to our department for primary infertility exploration.Prior semen analyses had demonstrated azoospermia.The patient had no particular pathological history.The physical examination revealed a well-masculinized body with normal hair density and distribution,without gynecomastia.However,genital examination revealed bilateral hypotrophic testes.

    • 摘要: The molecular control of osteoclast formation is still not clearly elucidated. Here, we show that a process of cell recognition mediated by Siglec15-TLR2 binding is indispensable and occurs prior to cell fusion in RANKL-mediated osteoclastogenesis. Siglec15 has been shown to regulate osteoclastic bone resorption. However, the receptor for Siglec15 has not been identified, and the signaling mechanism involving Siglec15 in osteoclast function remains unclear. We found that Siglec15 bound sialylated TLR2 as its receptor and that the binding of sialylated TLR2 to Siglec15 in macrophages committed to the osteoclast-lineage initiated cell fusion for osteoclast formation, in which sialic acid was transferred by the sialyltransferase ST3 Gal1. Interestingly, the expression of Siglec15 in macrophages was activated by M-CSF, whereas ST3 Gal1 expression was induced by RANKL. Both Siglec15-specific deletion in macrophages and intrafemoral injection of sialidase abrogated cell recognition and reduced subsequent cell fusion for the formation of osteoclasts, resulting in increased bone formation in mice. Thus, our results reveal that cell recognition mediated by the binding of sialylated TLR2 to Siglec15 initiates cell fusion for osteoclast formation.

    • 摘要: The effect of hydrogen injection on blast furnace operation and carbon dioxide emissions was simulated using a 1D steady-state zonal model.The maximum hydrogen injection rate was evaluated on the basis of the simulation of the vertical temperature pattern in the blast furnace with a focus on the thermal reserve zone.The effects of blast temperature and oxygen enrichment were also examined to estimate coke replacement ratio,productivity,hydrogen utilization efficiency,and carbon dioxide emission reduction.For blast temperature of 1200°C,the maximum hydrogen injection rate was 19.0 and 28.3 kg of H_(2)/t of hot metal(HM)for oxygen enrichment of 2vol%and 12vol%,respectively.Results showed a coke replacement ratio of 3-4 kg of coke/kg of H_(2),direct CO_(2) emission reduction of 10.2%-17.8%,and increased productivity by up to 13.7%depending on oxygen enrichment level.Increasing blast temperature further reduced the direct CO_(2) emissions.Hydrogen utilization degree reached the maximum of 0.52-0.54 H_(2)O/(H_(2)O+H_(2)).The decarbonization potential of hydrogen injection was estimated in the range from 9.4 t of CO_(2)/t of H_(2) to 9.7 t of CO_(2)/t of H_(2).For economic feasibility,hydrogen injection requires revolutionary progress in terms of low-cost H_(2) generation unless the technological change is motivated by the carbon emission cost.Hydrogen injection may unfavorably affect the radial temperature pattern of the raceway,which could be addressed by adopting appropriate injection techniques.

    • 摘要: Mesenchymal stem cells(MSCs)represent an important tool in veterinary regenerative medicine due to their ability to home to injury sites and secrete molecules that regulate niches into regenerative microenvironments.Successful cell therapy depends on many factors,including choice of administration route and application of understanding of cell potency and their therapeutic mechanisms.In this point of view,the authors leverage the tumultuous history of the field to demonstrate the need for clinicians to continually update themselves as new discoveries are made in order to avoid misalignments in the future,especially regarding administration routes and dose frequency,as well as to explore recent insights into MSC plasticity,therapeutic mechanisms,and cell delivery systems.
    • 摘要: 2022年5月11日,浙江大学医学院附属妇产科医院张丹教授团队在《美国妇产科杂志》(American Journal of Obstetrics and Gynecology)发表了题为“Embryo morphologic quality in relation to the metabolic and cognitive development of singletons conceived by in vitro fertilization/intracytoplasmic sperm injection:a matched cohort study”的研究论文(DOI:10.1016/j.ajog.2022.05.019),系统开展对出生子代的随访,发现新鲜卵裂期低质量胚胎移植后出生的单胎子代在4~6岁时的代谢和认知功能与高质量胚胎移植子代相当。

    • 摘要: We propose a new injection scheme that can generate electron beams with simultaneously a few permille energy spread, submillimeter milliradian emittance, and more than a 100 pC charge in laser wakefield accelerators. In this scheme, a relatively loosely focused laser pulse drivesthe plasma wakefield, and a tightly focused laser pulse with similar intensity triggers an interference ring pattern that creates onion-like multisheaths in the plasma wakefield. Owing to the change in wavefront curvature after the focal position of the tightly focused laser, the innermostsheath of the wakefield expands, which slows down the effective phase velocity of the wakefield and triggers injection of plasma electrons.Both quasicylindrical and fully three-dimensional particle-in-cell simulations confirm the generation of beams with the above mentionedproperties.

    • 摘要: BACKGROUND The incidence of infection with Mycobacterium abscessus(M.abscessus)has increased in recent years.This increase is partly associated with invasive cosmetic procedures.CASE SUMMARY The purpose of this case summary is to increase clinicians''awareness of M.abscessus infection and reduce mycobacterial infection caused by cosmetic procedures.We report the case of a 45-year-old woman who received acetyl hexapeptide-8(argireline)injections in the forehead and temples,and erythema,nodules,and abscesses appeared at the injection sites after one week.The pus specimens were examined by microbiological culture and confirmed to be positive for M.abscessus.Clarithromycin 500 mg twice daily and moxifloxacin 400 mg once daily were administered for 5 mo and the lesions gradually subsided.CONCLUSION We report here for the first time a case of infection with M.abscessus after argireline injection.This condition is easily misdiagnosed as a common bacterial infection.Microbiological examinations are helpful for diagnosis and standardized cosmetic procedures can prevent infection with M.abscessus.

    • 摘要: cqvip:Although mesenchymal stem cell-derived exosomes(MSC-exos)have been shown to have therapeutic effects in experimental periodontitis,their drawbacks,such as low yield and limited efficacy,have hampered their clinical application.These drawbacks can be largely reduced by replacing the traditional 2D culture system with a 3D system.However,the potential function of MSC-exos produced by 3D culture(3D-exos)in periodontitis remains elusive.This study showed that compared with MSC-exos generated via 2D culture(2D-exos),3D-exos showed enhanced anti-inflammatory effects in a ligature-induced model of periodontitis by restoring the reactive T helper 17(Th17)cell/Treg balance in inflamed periodontal tissues.Mechanistically,3D-exos exhibited greater enrichment of miR-1246,which can suppress the expression of Nfat5,a key factor that mediates Th17 cell polarization in a sequence-dependent manner.Furthermore,we found that recovery of the Th17 cell/Treg balance in the inflamed periodontium by the local injection of 3D-exos attenuated experimental colitis.Our study not only showed that by restoring the Th17 cell/Treg balance through the miR-1246/Nfat5 axis,the 3D culture system improved the function of MSC-exos in the treatment of periodontitis,but also it provided a basis for treating inflammatory bowel disease(IBD)by restoring immune responses in the inflamed periodontium.
