An E.coli strain which 'over-produces' L-tryptophan, comprises, in the genome thereof, a mutant gene which encodes partially-defective tryptophanyl-tRNA synthetase (preferably the trpS378 allele) - which when present in a non-producing strain, is characterized by a temperature-dependent tryptophan auxotrophy - and has a further mutation on the chromosome thereof in at least one gene selected from mtr (especially mtr - 24 ), aroP (especially aroP579 ), tnaA (especially tnaA1 ) and tnaB . The strains may be transformed with a plasmid which carries the tryptophan operon encoding an anthranilate synthase enzyme, freed from feedback inhibition by tryptophan, and/or which possess mutations in at least one gene selected from the trpEDCBA - group (which, when present in a non-producing cell, confers tryptophan auxotrophy thereon). L-tryptophan may be prepared by the cultivation of the said strains at ca. 30 degrees C (preferably in the presence of yeast extract), limiting growth by the concentration of available phosphate (in an appropriate medium), followed by fermentation (in the absence of yeast extract) at from 33 to 37 degrees C. The fermentation is preferably effected in the absence of antibiotics.
