The present invention describes a cost-effective, rapid, sensitive and specific diagnostic method of detecting Mycobacterium tuberculosis (MTB) direct in processed sputum based on complex that formed between carbol-fuchsin dye and mycolic acid solution with absorbance maxima ranging between 490-500 nm. To ensure validated detection of MTB in processed sputum containing mycolic acid was performed by using ATCC cultures of Mycobacterium species viz., Mycobacterium tuberculosis (ATCC 25177), M. avium subsp. avium serotype 1 (ATCC 15769), M. intracellular (ATCC 13950) and M. smegmatis (ATCC 14468).
