The object of the present invention is to provide 3-miRNAclassifiers of miR-3621 andmiR361-3p as a miRNA classifier for diagnosing lymph node metastasis in colorectal cancer. In addition, the present invention relates to a method for determining the occurrence of lymph node metastasis in colorectal cancer by using the miRNA classifier. Among 3-miRNA classifiers confirmed in the present invention, miR-342-3p and miR-361-3p are downregulated compared to the LNM-negative in LNM-positive CRC, and miR-3621 is upregulated compared to the LNM-negative in LNM-positive CRC, and thus, they may be utilized as biomarkers to determine the occurrence of lymph node metastasis in colorectal cancer. In addition, the expression of proteins expressed from target genes of the miR-342-3p, miR-361-3p, or miR-3621, namely E2F1, RAP2B, FOXM1, PIK3R3, AKT1, and WINT5A, is significantly increased as well in LNM-positive CRC, and thus, they may be utilized as a biomarker to determine the occurrence of lymph node metastasis in colorectal cancer. Accordingly, by using the proteins expressed from the 3-miRNA classifiers of the present invention and target genes thereof as a biomarker, it is possible to determine, in an accurate and rapid manner, whether lymph node metastasis has occurred in the early-stage colorectal cancer, and thus, the present invention may be effectively used to reduce the number of unnecessary surgeries in endoscopically resected T1-stage CRC.
