The present invention provides a miR-342-3p, miR-3621, and a 3-miRNA classifier of miR361-3p as a miRNA discriminator for diagnosing lymph node metastasis of colorectal cancer. Of the 3-miRNA classifiers identified in the present invention, miR-342-3p and miR-361-3p are down-regulated in LNM positive CRC compared to LNM negative, and miR-3621 is positive for lymph node metastasis. Lymph node metastasis is up-regulated compared to negative in CRC, which can be used as a biomarker for determining lymph node metastasis in colorectal cancer. In addition, E2F1, RAP2B, FOXM1, PIK3R3, AKT1, and WINT5A, which are proteins expressed from the target genes of miR-342-3p, miR-361-3p, or miR-3621, also significantly increased lymphocytic expression in CRCs. In addition, it can be used as a biomarker for determining lymph node metastasis in colorectal cancer.
