Pools of DNA from 20 buffalo and 20 cattle were compared for presence of Randomly Amplified Polymorphic DNA (RAPD) markers that might be useful in distinguishing between the two species. Using 10-decamer oligoneucliotide and an hour RAPD-PCR condition, primer scorable bands were produced. In addition, the market will be useful for exploration of the evolutionary relationship of the cattle and buffalo. The general approach used to identify species-specific DNA polymorphism has potentially broad application in definition of species will be generic value in studies of genome evaluation. It is concluded that RAPD polymorphism are useful as genetic markers for cattle and buffalo species differentiation.
