A lipase inhibitor from peel of cucumber: 1. purification and identification

Lipase Inhibitors can be used to prevent the development of off-flavour during food storage which may be caused by lipase, and also in pharmaceuticls for body weight control and so on. In the course of our screening test for lipase inhibitors form about 100 kinds of natural materials, we found that 70 percent methanol extract from peels of cucumber has a very potent inhibitory activity. An inhibitory active component in the extract was purifified and isolated by using liquid-liquid partition method, preparative thin layer chromatography, gel filtration chromatography and high performance liquid chromatography. The IC_50 of the active component was 0.0121 mg (28.5 nmol) for Rhizopus delemar lipase and the yield from 5 kg of cucumber was 11.9 mg. The chemical structure of this new lipase inhibitor was estimated to be 1-palmitoyl-2-lyso-sn-glycero-3-phosphatidyl methanol by the informations based on IR, NMR, and FAB-MS spectrometries.
