益寿丸对衰老小鼠脑线粒体DNA缺失突变的影响

         

摘要

目的:研究益寿丸对D-半乳糖( D-gal)所致衰老小鼠线粒体损伤的影响,探讨其抗衰老的作用机制。方法采用 PCR法,考察益寿丸对D-半乳糖致衰老模型小鼠脑线粒体DNA缺失突变的影响;并在透射电镜下对衰老动物脑组织线粒体的超微结构进行观察。结果模型组、益寿丸低剂量组及阳性药组小鼠海马mtDNA中存在着约3.87kb片断缺失。益寿丸高、中剂量组未见缺失片断。结论提示益寿丸有防御模型小鼠脑mtDNA缺失突变的作用,益寿丸在调节能量代谢方面具有一定改善作用。%OBJECTIVE To study the YiShou Pills on brain mitochondrial DNA missing of the aging model mice ,to explore the mechanism of its anti-aging.METHODS PCR was used to inspect the effect of YiShou Pills on brain mitochondrial DNA missing of the aging model mice induced by D-gal;And ultrastructure of aging mice brain mitochondria were observed under the transmission electron microscope.RESLUTS The results showed that model group,YiShou Pills low dosage group and positive controlled group carried 3.87kb mtDNA deficiency in brain of mice.While YiShou Pills high and middle dosage group could not carried mtDNA deficiency.CONCLUSION YiShou Pills could defense the brain mitochondrial DNA missing of the aging model mice ,which had the improvement action of regulating energy metabolism.
