鼻咽癌放射敏感性与MDR1基因多态性的相关性

         

摘要

目的 探讨鼻咽癌放射敏感性差异与多药耐药基因(MDR1)多态性的相关性.方法 选择48例行根治性放疗的鼻咽癌患者,根据放疗疗效分为抵抗组和敏感组,针对MDR1基因多态性(21外显子G2677T和26外显子C3435T)行PCR扩增、基因测序分型及构建G2677T-C3435T单体型.结果 G2677T TT基因型放射敏感性显著低于GG和GT基因型(P=0.017),C3435T TT基因型放射敏感性显著低于CC和CT基因型(P=0.022),携带2677T-3435T单体型的患者放射敏感性显著低于其他单体型携带者(P=0.013).结论 MDR1 G2677T和C3435T多态性可能提示鼻咽癌根治性放疗的疗效.%Objective To investigate the relevance between the polymorphisms of the human multidrug resistant gene ( MDR1 ) and radiosensitivity to radical radiotherapy of nasopharyngeal carcinoma(NPC). Methods Blood samples were collected from 48 NPC patients. The genotypes for MDR1 polymorphisms ( G2677T in exon 21 and C3435T in exon 26)were determined via the method of PCR and genotype sequencing, and the G2677T- C3435T haplotype was also constructed for analysis. Results The 2677TT genotypo was associated with a significantly lower radiasensitivity compared with the combined 2677GG and GT genotype( P =0.017). The 3435Tr genotypo was associated with a significantly worse radiotherapy response compared with the combined 3435CC and CT G enotypo(P = 0. 022). The haplotype of 2677T-3435T also indicated a significant decline of radiosensitivity compared with the other haplctypes (P = 0. 013). Conclusions The MDR1 G2677T and C3435T polymorphisms might help predict the response to radical radiotherapy as genetic markers in NPC patients.
