葛花提取液成分分析及其抑制小鼠体内酒精吸收实验

         

摘要

Objective To explore the main components of Pueravia Flowe extracted with two different solvents and the experiment effects in the inhibition of alcohol absorption in mice.Methods Pueravia Flowe was extracted with 95% ethyl alcohol and water respectively.The contents of four main components (i.e.,total alkaloids,flavonoids,reducing sugar and protein) in the extract were measured.Healthy mice were randomly divided into a blank group,a model control group,a positive control group,a Pueravia Flowe ethanol extract (low,medium and high doses) group and a Pueravia Flowe water extract (low,medium and high doses) group (n=20,respectively).Each drug administration group was administrated with corresponding drug by preventive gavage.30 minutes later,each group was administrated with liquor by gavage.Afterwards,half of the mice in each group were taken to record the disappearance time of righting reflex.The eyeballs of the other half of the mice were removed to collect blood.Their livers were also removed.Gas chromatographic method was taken to determine the ethanol concentration in mice plasm and microplate reader was used to determine the activity of alcohol dehydrogenase (ADH) in livers.Results The contents of total alkaloids,flavonoids,reducing sugar and protein in Pueravia Flowe alcohol extract were 8.1,179.2,134.0 and 0.9 mg/g respectively and 6.0,66.2,226.0 and 93.9 mg/g respectively in water extract.The disappearance time of righting reflex in drug administration team was longer than that in the model group,the plasma ethanol concentration was lower than that in the model group,and the liver ADH activity was higher than that in the model group;the disappearance time of righting reflex in the mice in Pueravia Flowe ethanol extract with the same dosage was shorter than that in the water extraction group,the ethanol concentration was greater than that in the water extraction group,and the liver ADH activity was higher than that in the water extract group.Conclusion After preventive gavage of mice,Pueravia Flowe ethanol extract and water extract can prolong the disappearance time of righting reflex in mice in a certain concentration range,lower the blood ethanol concentration in mice,improve the liver ADH activity in mice and prevent alcoholism.%目的 探讨两种不同溶剂提取葛花中主要成分及其抑制小鼠体内酒精吸收实验效果.方法 分别采用95%乙醇和水提取葛花,并测量提取液中总生物碱、总黄酮、还原糖、蛋白质4种主要成分的含量.健康小鼠随机分为空白组、模型对照组、阳性对照组、葛花醇提取物(低、中、高剂量)组、葛花水提取物(低、中、高剂量)组,每组20只.各给药组预防性灌胃给予相应的药物一次,30 min后各组灌胃给予白酒,随后各组随机取一半小鼠记录其翻正反射消失时间,另一半小鼠30 min后摘眼球取血,并摘取肝脏.气相色谱法测定小鼠血浆乙醇浓度,酶标仪测定肝脏中乙醇脱氢酶(ADH)活性.结果 葛花醇提液中总生物碱、总黄酮、还原糖、蛋白质的含量分别为8.1、179.2、134.0和0.9 mg/g,水提液中分别为6.0、66.2、226.0和93.9 mg/g.给药组小鼠的翻正反射消失时间均长于模型组,血浆乙醇浓度均低于模型组,肝脏ADH活性均高于模型组;同剂量的葛花醇提取物组小鼠的翻正反射消失时间短于水提取物组,乙醇浓度大于水提取物组,肝脏ADH活性高于水提取物组.结论 葛花醇提液和水提液经预防性灌胃小鼠后,在一定浓度范围内皆能延长小鼠翻正反射消失时间,降低小鼠血乙醇浓度,提高小鼠肝脏ADH的活性,从而预防酒精中毒.
