胰岛素诱导基因1和2在猪前体脂肪细胞分化过程的表达

摘要

为探讨胰岛素诱导基因1和2（Insig1，Insig2）在猪前体脂肪细胞中的表达规律，分离得到猪前体脂肪细胞，并诱导分化，利用油红O染色提取法测定了细胞中的甘油三酯含量，同时采用实时定量RT-PCR方法检测了脂肪细胞分化标志基因、胰岛素诱导基因1和2的表达。结果表明：在猪前体脂肪细胞诱导分化过程中，Insig1的mRNA表达显著增加（P＜0.05）；Insig2的mRNA表达量在诱导后24 h显著增加（P＜0.05），而后降低。%In order to investigate the expression of insulin induced genes 1 and 2 during porcine preadipocyte differentiation process,porcine preadipocyte was isolated and introduced .The concentration of cell triglyceride was measured by Oil Red O staining .Real-time RT-PCR method was applied to in-vestigate mRNA expression level of the adipocyte differentiation marker genes,insulin induced genes 1 and 2 .During porcine preadipocytes differentiation,Insig1 mRNA expression increased significantly (P<0.05);Insig2 mRNA expression increased obviously (P<0.05)at 24 h after induction,then de-creased .The research results will provide a theoretical basis for the further study of fat metabolism and pork quality trait improvement .
