山楂叶总黄酮对脊髓损伤大鼠运动功能和炎症反应的影响

     

摘要

Objective:To investigate the effect of hawthorn leaves flavonoids on motor function and inflammatory reaction in rats with spinal cord injury(SCI).Methods:Thirty SD rats were randomly divided into sham-operated group,SCI group and hawthorn leaves flavonoids low-dose(5mg/kg) ,middle-dose(10mg/ kg) and high-dose(20mg/kg) groups,with 6 rats in each group.Except for the sham-operated group,the rat SCI model was established by Allen's method.The motor function were evaluated by Basso-Beattie-Bresnahan(BBB) score.The levels of interleukin(IL) -1β,IL-10 and tumor necrosis factor(TNF) -αwere detected by enzyme linked immunosorbent assay(ELISA).Results:Compared with sham-operated group, the score of BBB in SCI group was significantly decreased,while the levels of IL-1β,IL-10and TNF-αin spinal cord tissues were increased(P<0.05). The concentrations of IL-1β,IL-10and TNF-αin spinal cord tissues of the three treatment groups decreased with the increase of the dose of hawthorn leaves flavonoids(P<0.05).Conclusion:Hawthorn leaves flavonoids could inhibit inflammatory response and improve motor function in SCI rats.%目的:探讨山楂叶总黄酮对脊髓损伤(SCI)大鼠运动功能及炎症反应的影响.方法:选取30只SD大鼠,随机分为假手术组(A组)、SCI组(B组)及山楂叶总黄酮低剂量(5mg/kg)组(C)、中剂量(10mg/kg)组(D组)和高剂量(20mg/kg)组(E组),每组6只.除A组外,其他各组均采用改良Allen''s法复制SCI模型.于SCI后24h、48h、72h,采用Basso-Beattie-Bresnahan(BBB)评估大鼠的运动功能.采用酶联免疫吸附试验(ELISA)法测定脊髓组织中白介素(IL)-1β、IL-10和肿瘤坏死因子(TNF)-α水平.结果:与A组比较,B组BBB评分明显降低,脊髓组织IL-1β、IL-10和TNF-α水平显著升高(P<0.05);随着山楂叶总黄酮浓度的增加,BBB评分逐渐升高,IL-1β、IL-10和TNF-α浓度下降,且与B组比较差异均有统计学意义(均P<0.05).结论:山楂叶总黄酮能够有效抑制SCI大鼠炎症反应,改善肢体的运动功能.
