HPLC-ELSD法测定昆仑雪菊中氨基酸种类

摘要

为了更好的开发、利用昆仑雪菊，研究昆仑雪菊中的氨基酸种类，建立一种高效液相色谱法-蒸发光散射检测器（HPLC-ELSD）直接测定昆仑雪菊中氨基酸种类的分析方法。以PrevailTM C18（4.6 mm×250 mm，5μm）色谱柱、梯度洗脱、漂移管温度110℃、氮气流量2.5 L/min为色谱条件，在40 min内完成20种氨基酸的检测。该方法能够简便、准确地测定昆仑雪菊中未水解氨基酸的种类，同时为昆仑雪菊中氨基酸的提取提供依据。%In order to explore and utilize the Coreopsis tinctoria flower more efficiently,a high performance chromatograph evaporative light-scattering detection method for the determination of amino acids was studied. The assay was conducted on PrevailTM C18(4.6 mm×250 mm,5μm) column,gradient elution,and the detection was performed at the temperature of drift tube of 110℃and the nitrogen gas flow rate of 2.5 L/min. Within 40 minutes, 20 kinds of amino acids were detected. This method was simple,accurate to direct determination of amino acids in Coreopsis tinctoria flower,which provided information to extract amino acids in Coreopsis tinctoria flower.
