防风通圣丸中黄芩最佳提取工艺的优选研究

         

摘要

Objective To determine the optimal extraction solvent and extraction technology of scutellaria in Fangfeng Tongsheng Pills. Methods HPLC method was adopted with Welchrom- C18(250 mm×4.6 mm, 5 μm) column. The mobile phase consisted of 0.4%phosphoric acid-methanol (47:53). The flow rate was 1.0 mL/min, detection wavelength was 280 nm and the column temperature was 25℃. With baicalin as evaluation index, the content of baicalin in decoction samples, alcohol extraction samples, samples of separate decoction and mixed decoction was determined. Then, the extraction yield was calculated to obtain the optimal extraction technology of scutellaria in Fangfeng Tongsheng Pills. Results The extraction yields of baicalin in decoction samples and alcohol extraction samples were 72.70%and 85.66%. The extraction yields of baicalin in samples of separate decoction and mixed decoction were 75.13 % and 70.14 %. Conclusion Combined with the industrial production factors, the optimal extraction technology of scutellaria in Fangfeng Tongsheng Pills can be mixed-decoction of scutellaria, gypsum and talcum powder, which can provide the experimental basis for dosage form reform.%目的:确定防风通圣丸中适宜提取黄芩的溶剂及最佳工艺。方法用高效液相色谱法,色谱柱Welchrom-C18柱(250 mm×4.6 mm,5μm);流动相0.4%磷酸-甲醇(47:53);检测波长280 nm,流速1.0 mL/min,柱温25℃;以黄芩苷为评价指标,通过测定黄芩水煎和醇提以及单煎和合煎样品中黄芩苷含量及计算提取率,确定黄芩的最佳提取工艺。结果黄芩水煎和乙醇提取样品中黄芩苷提取率分别为72.70%和85.66%;黄芩单煎和与矿物质合煎样品中黄芩苷的提取率分别为75.13%和70.14%。结论结合工业生产的实际和经济因素,确定防风通圣丸中黄芩药材的最佳提取工艺为黄芩、石膏、滑石三者合并水煎,为改革防风通圣丸的剂型提供了试验依据。
