Objective To develop an HPLC method for simultaneous determination of two oleanane type aglycones (gypsogenin and quillaic acid) in Gypsophila oldhamaina Miq..Methods The gypsogenin and quillaic acid in Gypsophila oldhamiana Miq. after acid hydrolysis were determined by HPLC. Agilent Eclipes XDB-C18column (250 mm×4.6 mm, 5μm) was adopted, using methanol-acetonitrile-0.1% phosphoric acid solution (51:9:40) as mobile phase at the flow rate of 1.2 mL/min. The column temperature was 25℃, the detection wavelength was 210 nm, and the injection volume was 10μL.Results The calibration curve of gypsogenin was in good linearity at 2.920-17.52μg (r=0.9997) and the average recovery was 99.3 %, with RSD of 0.82 %. The calibration curve of quillaic acid was in good linearity at 8.040-48.24μg (r=0.9998) and the average recovery was 98.5 %, with RSD of 1.2 %.Conclusion This method is simple, accurate and reproducible, and can be used to determine the content of gypsogenin and quillaic acid in Gypsophila oldhamiana Miq. simultaneously.%目的 建立高效液相色谱法(HPLC)同时测定丝石竹药材中丝石竹皂苷元和皂皮酸两个齐墩果烷型苷元含量的方法.方法 丝石竹药材经酸水解后,采用HPLC测定丝石竹皂苷元和皂皮酸含量.采用Agilent Eclipse XDB-C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇-乙腈-0.1%磷酸水溶液(51:9:40)为流动相,流速1.2 mL/min,检测波长210 nm,柱温25℃,进样量10μL.结果 丝石竹皂苷元和皂皮酸进样量分别在2.920~17.52μg(r=0.9997)和8.040~48.24μg(r=0.9998)范围内与峰面积呈良好的线性关系.两者平均回收率分别为99.3%(RSD=0.82%)和98.5%(RSD=1.2%).结论 该方法简便、准确、重复性强,可作为同时测定丝石竹药材中丝石竹皂苷元和皂皮酸含量的方法.
