和胃安神方对失眠大鼠脑组织谷氨酸与γ-氨基丁酸含量的影响

         

摘要

目的 观察和胃安神方对失眠大鼠脑组织谷氨酸(GLU)、γ-氨基丁酸(GABA)含量的影响,探讨其治疗失眠的作用机制.方法 将40只雄性Wistar大鼠随机分为正常组、模型组、和胃安神方组及地西泮组,每组10只.除正常组外,其余各组大鼠均用对氯苯丙氨酸建立大鼠失眠模型.和胃安神方组和地西泮组分别给予和胃安神方和地西泮水溶液灌胃,其余2组给予生理盐水灌胃.6d后,断头处死并冰台上分离大鼠海马及下丘脑,低温离心后分离上清液,高效液相色谱法测定GLU、GABA含量.结果 与正常组比较,模型组大鼠海马及下丘脑GLU含量显著升高(P<0.01),GABA含量显著下降(P<0.01);与模型组比较,和胃安神方组大鼠海马及下丘脑GLU含量显著下降(P<0.01),GABA含量显著升高(P<0.01).结论 和胃安神方可以调节失眠大鼠脑内的氨基酸类神经递质GLU及GABA含量,从而发挥治疗失眠的作用.%Objective To study the effect of Hewei Anshen Decoction on cerebral contents of glutamic acid (GLU) and garnma-aminobutyric acid (GABA) in insomnic rats and explore its mechanism in treating insomnia. Method Forty healthy male Wistar rats were randomly divided into normal group, model group, Hewei Anshen Decoction group and diazepam group, with 10 rats in each group. All rats but those in the normal group were intraperitoneally injected with PCPA to make model of insomnia. Hewei Anshen Decoction group were treated with Hewei Anshen Decoction, diazepam group with diazepam solution, and the other two groups with normal saline for 6 days. After that, rats were killed, hippocampus and hypothalamus were removed and the contents of GLU and GABA were analyzed using high performance liquid chromatography (HPLC). Result Compared with normal group, GLU levels in hippocampus and hypothalamus of model group were significantly higher, while GABA levels significantly lower (P<0.01). Compared with model group, GLU levels in hippocampus and hypothalamus of Hewei Anshen Decoction group were significantly lower, and GABA levels significantly higher (P<0.01). Conclusion Hewei Anshen Decoction can treat insomnia through regulating central neurotransmitters GLU and GABA of insomnic rats.
