The role of rapid, nongenomic actions of aldosterone in acute cardiovascular events has receivedparticular attention. Our aim was to study the effect of aldosterone on thrombus formation in adrenalectomizedrats with streptozotocin-induced diabetes and the role of mineralocorticoid (MR) and glucocorticoid (GR)receptors in hormone action. Arterial thrombosis was induced by electrical stimulation of the carotid artery.Marked increase in the thrombus weight and shortening of the time to the artery occlusion were observed duringacute aldosterone infusion. Eplerenon, MR antagonist or RU-486, GR antagonist markedly prolonged the time toocclusion and reduced the rate of thrombosis. Our data have shown prothrombotic effect of aldosterone inadrenalectomized, diabetic rats which seems to involve both MR and GR dependent pathways.
