Thiol-Specific Nitroxides for Site Directed Spin Labeling and Measurement of Thiol Redox State

The traditional thiol-specific reagents include such reactive functionalities as maleimides, iodoacetates, organomercurials, alkylthiosulfonates, and disulfides. The alkylthiosulfonate and biradical disulfide spin labels are distinguished by their high selectivity for sulfhydryl groups, rapid reactivity under mild conditions without requiring a large excess of reagent, and the general reversibility of the formation of disulfide bonds upon the addition of thiols such a beta-mercaptoethanol or dithiothreitol.
