Mimotope Analysis of Human Single Chain Antibodies Cross-reacted to Both H1(H1N1) and H5(H5N1) Subtype Influenza Viruses

Recently, we have established twenty-seven single chain antibody (scFv) clones which are specific to influenza H5 hemagglutinin (HA), from human scFv displaying phage library using recombinant H5 fused with immunogloblulin Fc (H5-Fc). One of these antibodies (2A1) cross-reacted to both H1 and H5 influenza vaccine strain in ELISA assay. In this paper, we analysed the mimotope of 2A1 scFv using peptide-displaying phage library, Ph.D-12. and found a sequence homology in the native H5 and H1 sequence.
