1. A method was developed to co-register and compare MALDI images obtained from serial tissue sections. 2. Alignment was estimated to be accurate to +/- one pixel (60 μm for the data shown here) 3. Images could be aligned across different MS methods on consecutive sections, e.g. MS data acquired in +ve ion mode were matched with MS/MS data acquired in -ve ion mode. This is critical as it permits comparison of the spatial distribution between different lipid species. 4. Image correlation from serial sections for GC/GC images and BMP/BMP images were stronger than for GC/BMP images. 5. Future work: evaluation of alternative image correlation algorithms.
