Chemical cross-linking and high-resolution mass spectrometry were used to identify the interaction site of CusF/CusB during metal transfer in vitro. These results further confirm that the interaction between CusF and CusB is metal-dependent. The crosslinks demonstrate an interaction site that includes the metal-bound regions of both proteins. These results support the role of CusF as a metallochaperone and further confirm the N-terminal metal binding site of CusB.
