Changing the analyte ionization state by adjusting the pH of the extracts prior to evaporation, modify the extraction mode or the evaporator model/brand can be very useful to reduce contamination. Lower temperature and minimum N2 gas flow rate should be applied when performing evaporation of small molecules. A new automated liquid handling cross-well contamination test was added in all validation procedures using automated platforms to control such issues. This test is to perform a checkerboard of blanks and high concentration and to alternate for a minimum of 3 consecutive rows on a 96-well plate.
