Aims: In this study, the production of pre-amplified ROC23 and ROC25 with primers combinations E£A/M£C was diluted as DNA template to filtrate the 110 primer combinations. Methods and Results: The genomic DNA of the ROC25 and ROC23 were analyzedwith the technology of Amplified Fragment Length Polymorphism (AFLP). Five pairs of AFLP primers were selected from 110 pairs, they are E_2/M_4, E_4/M_5, E_5/M_6, E_3/M_7 and E_2/M_6. Significance of Study: The five pair of AFLP primers showed rich polymorphism and strong resolution capacity of the genomic DNA of sugarcane, which will provide reference for sugarcane genetic research with AFLP in the future.
