Identification of differentially expressed genes during disease resistance response from rice by cDNA arrays

A normalized cDNA library was constructed with 14 different tissues from rice variety Minghui 63, including tissues after inoculation with the pathogens Xantfiomonas olyzae pv. oryzae (Xoo) and Pyricularia grisea. This library contains about 62,000 clones with an average insert length of 1.2 kb. Approximately 20,000 clones from the library were arrayed on nylon film. Total RNA samples, isolated from different bacterial blight or blast resistance varieties and near-isogenic lines induced with different strains or isolates of Xoo or R grisea, respectively, were used as probes to screen the cDNA arrays for identification of defense-related genes. Some 110 differentially expressed sequences were identified. The expression of 56 of these sequences increased and 54 decreased or was inhibited after inoculation with Xoo or P. grisea. Sequence analysis of the 56 sequences showing increased expression after pathogen induction reveals that about two-thirds of these sequences match with known functional proteins in the databases, which can be classified into several groups according to the putative functions. The cDNA array technique can provide an efficient tool for identifying defense-related genes.
