ANTIBODY CAPABLE OF RECOGNIZING HUMAN-DERIVED CYTOCHROME P4501A1

PURPOSE: To obtain a new antibody useful, for e.g. accurately and easily quantifying cytochrome P4501A1 as an oxidative metabolic enzyme, by separation/ purification from the mammal blood immunosensitized with recombinant human- derived cytochrome P4501A1 manifested in yeast. ;CONSTITUTION: A human liver-derived cDNA library is cloned by the PCR technique with a primer for cloning human-derived cytochrome P4501A1 gene, the resultant human-derived cytochrome P4501A1 gene is then incorporated into a vector and transfected into host cells such as yeast to effect transformation. The resultant transformant is cultured to obtain recombinant human-derived cytochrome 4501A1. Next, a kind of mammal such as rabbit is administered with the recombinant human-derived cytochrome 4501A1 as antigen to immunosensitize the rabbit, from which the blood is extracted and then centrifuged to gather antiserum, from which an antibody is separated and purified, thus obtaining the objective new antibody presenting no cross reactivity to the other human-derived cytochrome P450 molecular kinds and capable of specifically recognizing human-derived cytochrome P4501A1.;COPYRIGHT: (C)1996,JPO
