PROBLEM TO BE SOLVED: To enable analysis of a diluted horseradish peroxidase enzyme(HRP) and to measure the subject intermediate in high sensitivity by oxidizing a deferrioxamin-containing reagent with a reaction intermediate of HRP and H2O2 in the presence of both HRP and H2O2 and measuring the electron spin resonance of a deferrioxamin nitroxide radical. ;SOLUTION: In this method for measuring a reaction intermediate of a peroxidase enzyme (e.g. horseradish peroxidase enzyme) and hydrogen peroxide, a reagent containing deferrioxamin of the formula is oxidized with a reaction intermediate a hydrogen peroxide and the peroxidase enzyme in the presence of the peroxidase enzyme and hydrogen peroxide to form a deferrioxamin nitroxide radical and the electron spin resonance of the deferrioxamin nitroxide radical is measured. Consequently, the analysis of diluted peroxidase enzyme which could not be measured by a conventional method can be measured and the reaction intermediate of the objective peroxidase enzyme and hydrogen peroxide is measured in high sensitivity.;COPYRIGHT: (C)1997,JPO
