STRUCTURAL FEATURE OF CONJUGATE OF UBIQUITIN-BOUND DOMAIN OF YEAST DSK2 WITH MONOUBIQUITIN AND MECHANISM OF RECOGNIZING MONOUBIQUITIN BY UBIQUITIN-BOUND DOMAIN OF YEAST DSK2
PROBLEM TO BE SOLVED: To carry out an analysis of a mechanism of recognizing ubiquitin of a ubiquitin-bound domain at an atomic level.;SOLUTION: A method for predicting a three-dimensional structure of a conjugate of a ubiquitin-bound domain with the ubiquitin comprises the following steps: (i) a step of preparing the conjugate of the ubiquitin-bound domain of a yeast DSK2 with monoubiquitin or a three-dimensional structure of a part thereof specified by atomic coordinates in which the root mean square deviation related to the distance from the three-dimensional structure specified by the atomic coordinates of Table A is 0.34 for atoms constituting the main chain and 0.99 for all the atoms in the ubiquitin-bound domain which is a region of 328-373 residue numbers on the C-terminal side of the yeast DSK2 and in a region of 1-71 residue numbers of the monoubiquitin and (ii) a step of constructing the three-dimensional structure of the conjugate of a protein containing the ubiquitin-bound domain with the ubiquitin by a homology modeling using the three-dimensional structure prepared in the step (i) as a template.;COPYRIGHT: (C)2006,JPO&NCIPI
