A method for preparing a polysaccharide of Schizophyllum commune is provided to produce the desired polysaccharide with high yield at relatively low cost within short period of time and obtain a transparent culture solution by maintaining the concentration of hypha low and increasing the production amount of the polysaccharide of Schizophyllum commune, which is extracellularly secreted, thereby the obtained polysaccharide being used as a raw material of a functional food and a cosmetics without further purification. A method for preparing a polysaccharide of Schizophyllum commune comprises the steps of: (a) after selecting a strain with excellent growth speed and excellent Schizophyllum commune polysaccharide productivity from harvested Schizophyllum commune, culturing the selected strain in a potato dextrose agar(PDA) culture medium; (b) after cutting the cultured strain into a size of 3mm, shake-culturing the 5-10 fragments in 100ml of a base culture medium consisting of 4 wt.% of white sugar, 0.09 wt.% of an extract of yeast and 95.91 wt.% of distilled water and contained in a flask at a temperature of 25 deg.C for 3-4 days with the speed of 150 rpm; (c) aseptically homogenizing the solution obtained from the step(b) using a homogenizer with 15,000-20,000rpm for 1 minute; (d) culturing 0.2-0.8 vol/vol.% of the homogenized solution as a culture inoculating source in a sterile culture medium at a temperature of 15-30 deg.C for 4-5 days using a 20 liter volume water bottle or an air-lift incubator with only supplying compressed air thereto, wherein the sterile culture medium comprises 2-7 wt.% of white sugar, 0.02-0.09 wt.% of an extract of yeast and the remaining amount of distilled water, the pH of the sterile culture medium is not adjusted and an anti-foam is no added; (e) vacuum-filtering the culture solution obtained from the step(d) to effectively separate hypha from the culture filtrate; and (f) adding an excipient and a preservative to the isolated culture filtrate and then packing it.
