Method for preparing a chemically modified protein (P1) comprises (a) modifying an auxiliary protein (AP), containing a reactive functional group and comprising intein fragment (IF) and extein fragment (EF), and containing the group (G) that is to be introduced into the target protein (TP); and (b) reacting TP, which is to be modified and is fused to a complementary IF, such that modified EF becomes bound to TP. An independent claim is also included for pharmaceuticals and diagnostic agents that contain (P1) made by the new method.
