The present invention relates to a method and a device for nanopore based single-molecule protein/protein interaction detection. The invention combines the use of a charged polymer to drive a target molecule to the entrance of a nanopore, for the charged polymer to thread through the pore, and the molecule attached to the polymer to be held at the entrance of the nanopore as a dumbbell. Trans-membrane ionic current measurements can be used to monitor the ion conductance of a single nanopore. Depending on the pore dimensions, single molecules can be detected in this way.
