PROBLEM TO BE SOLVED: To provide polyamide-bonded stationary phases for hydrophilic interaction chromatography (HILIC) and new HILIC method for use in characterizing giant biomolecules that are modified with polar groups and are known in the art as glycan.SOLUTION: The present invention provides particularly novel, polyamide-bonded materials that are designed for efficient separation of giant biomolecules, for example, materials having large pores (wide pores) at a high rate. Furthermore, the present invention advantageously provides new HILIC methods that can be used in combination with the materials of the stationary phase described herein to effectively separate the glycoform of proteins and peptides by eliminating the earlier unsolved problems, such as aggregation of protein samples on the column, low sensitivity of chromatographic detection of glycan portion, and low resolution of peaks due to limited pore diffusion and long diffusion paths within/between particles.
