The present invention relates to a method of preparing multi-layer transfersomes containing linoleic acid and alpha-linoleic acid ester complexes using the hydrolysis of flax seed oils-derived organic acid and the esterification of fatty acids. More specifically, the method comprises: an organic acid hydrolysis step; a glycerin removal step; a fatty acid esterification step; a degassing and filtration step; a complex lipid preparation step; an aqueous chemical solution preparation step; a transfersome preparation step; and a transfersome microparticulation step. According to the present invention, the method increases the reaction speed and reaction conversion rate, has a fast emulsification speed, and exhibits emulsion stability, and thus has the effect of increasing the transdermal absorption rate.;COPYRIGHT KIPO 2019
