Preclinical Toxicologic Evaluation of Bruceantin (NSC-165,563) in Dogs

The infusion of Bruceantin as single dose of 1.25 (0.0625) or 0.312 (0.0156) mg/sq. m. (mg/kg) or as five daily doses of 0.626 (0.0313) mg/sq. m./day (mg/kg/day) was not toxic to dogs. Though traces of occult fecal blood were observed after treatment, evidence in dicated that this observation was a false positive and reflected the presence of peroxidase-like activity from sources other than blood. A few hematologic changes (leukopenia, thrombocytopenia, and anemia) were also noted, but none appeared to be related to treatment. These studies support the authors previous conclusion that 1.25 mg/sq. m./day is the HNTD (highest nontoxic dose) for single or five daily doses of Bruceantin.
