In the present study, a successful attempt was made to establish the aseptic culture of Cinnamomum tamala Nees from nodal explants and to evaluate the factors influencing in vitro morphogenesis. About 45.6% nodal explants collected during July established successfully on full strength MS medium fortified with sucrose (3%, w/v), polyvinyl pyrollidone (PVP; 100 mg L-1) and benzyl adenine (BA; 9 aM), where as many as 5.4 meristematic loci were invoked after 5-6 wk of culture initiation. The shoot buds developed on initiation medium converted into microshoots on transferring to MS medium conjunct with sucrose (3%) and lcinetin (Kn; 3 mu M). The inicroshoots were rooted by culture on MS medium fortified with sucrose (3%) and alpha-naphthalene acetic acid (NAA; 3 mu M), where as many as 10 roots developed per microshoot. The rooted plantlets were transferred to potting mix containing soil, sand and decayed wood powder at 1:1:1 ratio and maintained in the polyhouse at ca 75% shading of light for 2 months. The acclimatized plantlets were transferred to the nature and registered 70% survival after 2 months of transfer.
