Pharmacokinetics of topiramate in healthy Chinese volunteers

To study the pharmacokinetic profile of a single dose of topiramate in healthy Chinese volunteers. Methods Topiramate was administered to subjects as a single oral dose of lOOmg. Model-independent pharmacokinetic data analysis was performed on plasma concentration data. Results Maximum plasma concentration (C_(max)) was observed 1.22 hours after administration. Mean values for plasma C_(max) and area under the concentration-time curve (AUC) was 1.27 mugcentre not mL~(-1) and 46.9 mugcentre not hcentre not mL~(-1), respectively. Mean oral clearance (CL/F) was 36.0 mLcentre not min~(-1) and mean volume of distribution (V_d) was 110.6 L. Elimination half-life (t_(1/2)) value was calculated to be 35.5 hrs. Conclusion The values of AUC and CL/F obtained here were similar to those of the western counterparts reported, t_(max) was slightly shorter, but C_(max) was prominently lower, while t_(1/2) and Vd were notably higher. These results offered relevant information for rational use of topiramate in clinical practices with Chinese people.
