Metabolomics of the Effect of 1-MCP and CA during Ripening and Storage in Apples

Fruit ripening involves complex structural and biochemical changes comprising changes in a large number of metabolites. 1-methylcyclopropene (1-MCP) affects metabolic changes related to ripening by blocking the ethylene receptors, thereby inhibiting ethylene production. Metabolic profiling could provide useful insight into the metabolic regulation underlying fruit ripening and responses to environmental perturbations. To this end, we profiled dynamic changes in the metabolome of 'Jonagold' apple associated with 1-MCP treatment. 1-MCP reduced ethylene production and respiration rate during ripening at 18°C, RA storage at 1°C and controlled atmosphere (CA) storage (1% O2, 3% CO_2). Partial least square discriminant analysis (PLS-DA) on the metabolicprofiles of 1-MCP treated and control apples revealed metabolic divergence in ethylene, organic acid, sugar and amino acids metabolism distinctively attributed to each group. During ripening at 18°C, isoleucine, valine, aspartic acid, glycine, serine and glucose were highly positively associated with 1-MCP treatment, whereas 1-Aminocyclopropane-l-carboxylic acid (ACC) was highly positively correlated with untreated fruit.
