Root segments derived from cultures of A. kolomikta were cultured in vitro on BW medium supplemented with 2,4-D (0, 0.1, 1 or 10μM) and zeatin (0, 1 or 10μM) at 25deg C, under a 16-h photoperiod with a light intensity of 70μmol m-2 s-1. Calluses were formed at all concentrations of 2,4-D and zeatin. Of the adventitious buds which formed from callus, 92% developed into shoots in the presence of 2,4-D at 0.1μM + zeatin at 10μM. Root segments excised from cultures formed the most somatic embryos when cultured with 2,4-D and zeatin at 10μM. When young leaflets with roots were cultured on BW medium with zeatin, adventitious buds formed on the root tip. The rate of adventitious bud formation increased with increasing zeatin concentration. Adventitious buds developed into shoots and shoots rooted and grew into vigorous plantlets when transferred to Miller's medium supplemented with NAA at 1μM.
