The studies discussed below investigated the release of ketoprofen (formulated as an oral solution) from liquid feed. According to the label claim, this preparation has to be administered via drinking water Today, many pigs are raised using pulp or liquid feeding, and their need for drinking water is mainly supplied through feeding. The relative bioavailability of ketoprofen after administration in liquid feed was therefore examined in three studies: 1. Release of ketoprofen from liquid feed compared to the stock solution of the preparation using water of varying quality (in vitro), 2. Recovery of ketoprofen from the liquid and the solid phases of liquid feed (in vitro), 3. Bioavalability after administration as an oral bolus with drinking water compared to liquid feed (in vivo).The rate of recovery of ketoprofen from liquid feed (87%) was comparable to the recovery from pure water (92 %). In the liquid phase of the food samples, the content of ketoprofen was 26% to 46% of the initial amount, the other portion of ketoprofen remaining in the solid phase. Water hardness, water temperature und type of feed had no influence on the rate of recovery. Bioavailability and maximum concentration of ketoprofen were higher in pigs after administration via liquid feed compared to administration via drinking water (C-max: 3.33 vs. 3.01 mg/L; AUC: 11.95 vs. 10.55 mg*hr/L). The results show that ketoprofen reversibly binds to feed particles. However, bioavailability from liquid feed preparations proved to be at least as high as from drinking water under the test conditions of these studies.
