ThestructureofGE82832,atranslocationinhibitorproducedbyasoilmicroorganism,isshowntobehighlyrelatedtothatofdityromycin,abicyclodecadepsipeptideantibioticdiscoveredlongagowhosecharacterizationhadneverbeenpursuedbeyonditsstructuralelucidation.GE82832anddityromycinwereshowntointerferewithbothaminoacyl‐tRNAandmRNAmovementandwiththePireleaseoccurringafterribosome‐andEF‐G‐dependentGTPhydrolysis.ThesefindingsandtheunusualribosomallocalizationofGE82832/dityromycinnearproteinS13suggestthatthemechanismofinhibitionentailsaninterferencewiththerotationofthe30Ssubunit“head”whichaccompaniestheribosome‐unlockingstepoftranslocation...
