宫颈癌组织中受体相互作用丝氨酸/苏氨酸蛋白激酶4高表达与宫颈癌预后的相关性分析

Objective To investigate the expression of receptor interaction serine/threonine protein kinase 4 (RIPK4)in cervical cancer and its correlation with the clinicopathological parameters of cervical cancer.Methods The subjects were 39 patients with cervical cancer,and we collected all the cancer tissues and normal tissues adja-cent to the cancer of the cervix.Real-time quantitative PCR was used to detect the expression of RIPK 4 mRNA in cervical cancer tissues and normal cervix tissues.The expression of RIPK4 protein in cervical cancer tissues and normal cervix tissues was detected by immunohistochemistry.The correlation between the expression of RIPK4 in cervical cancer and the clinicopathological parameters of cervical cancer were analyzed.Results RIPK4 mRNA ex-pression in normal tissues is 1.5 ± 0.8;RIPK4 mRNA expression level in cervical cancer is 4.3 ± 1.6,the expres-sion level of RIPK4 mRNA in cervical cancer was significantly increased,there were significant differences(P<0.05).RIPK4 protein was negative in normal cervical tissues.RIPK4 protein was positive in cervical cancer tis-sues.14 cases(36%)of cervical cancer patients had weakly positive expression of RIPK 4 protein,and 25 cases (64%)had strongly positive expression of RIPK4 protein in cervical cancer patients.Statistical analysis showed that FIGO stage and lymph node metastasis were significantly associated with high expression of RIPK 4 in cervical cancer tissues(P<0.05).However,age,histological grade,and tumor size were not associated with higher ex-pression of RIPK4(all P > 0.05).Conclusion Statistical analyses indicated that FIGO stage and lymph node me-tastasis were significantly associated with higher expression of RIPK 4.%目的 探讨受体相互作用丝氨酸/苏氨酸蛋白激酶4(RIPK4)在宫颈癌组织中表达水平及与宫颈癌临床病理参数的相关性.方法 研究对象是39例宫颈癌患者,收集所有宫颈癌患者癌组织和癌旁正常组织.应用实时定量聚合酶链反应(PCR)方法检测宫颈癌组织及正常宫颈组织中RIPK4 mRNA表达水平.应用免疫组织化学方法检测宫颈癌组织及正常宫颈组织中RIPK4蛋白表达水平.分析宫颈癌组织中RIPK4表达与宫颈癌临床病理参数相关性.结果 正常组织中 RIPK4 mRNA表达水平是1.5 ± 0.8;宫颈癌组织中 RIPK4 mRNA表达水平是4.3 ± 1.6,宫颈癌组织中RIPK4 mRNA表达水平显著增高(P<0.05).RIPK4蛋白在正常宫颈组织中表达阴性;RIPK4蛋白在宫颈癌组织中表达阳性,其中14例(36%)宫颈癌患者RIPK4蛋白表达弱阳性,25例(64%)宫颈癌患者RIPK4蛋白表达强阳性.统计分析显示,国际妇产科联盟(FIGO)分期和淋巴结转移与宫颈癌组织中RIPK4高表达显著相关(P <0.05).结论 宫颈癌组织中RIPK4高表达与FIGO分期和淋巴结转移显著相关.
