新辅助化疗对子宫颈癌组织中NF-κB、LRP mRNA和蛋白表达的影响

         

摘要

目的 观察新辅助化疗(NACT)前后子宫颈癌组织中NF-κB、LRP mRNA和蛋白的表达变化,并探讨其临床意义.方法 采用RT-PCR和免疫组化法检测40例子宫颈癌患者NACT前后癌组织中的NF-κB、LRP mRNA和蛋白.结果 NACT后子宫颈癌组织中NF-κB、LRP mRNA表达量分别为67.595±0.914 、66.912±0.752,明显高于NACT前的50.340±0.780、50.048±0.770,P均<0.05.NACT后子宫颈癌组织中NF-κB、LRP蛋白阳性表达率分别为80.0%、92.5%,明显高于治疗前的50.0%和62.5%,P均<0.05.化疗前后肿瘤组织中的NF-κB、LRP蛋白表达均呈正相关(r分别为0.389、0.379,P均<0.05).结论 NACT后子宫颈癌组织中的NF-κB、LRP mRNA和蛋白较化疗前均明显升高(P均<0.01).NF-κB与LRP可能参加了子宫颈癌多药耐药的发生.NACT前联合检测子宫颈癌组织中的NF-κB、LRP mRNA和蛋白有助于评估预后及避免多药耐药.%Objective To investigate relationship between the expression of NF-κB and LRP mRNA and protein in cervicis carcinoma tissue before and after neoadjuvant chemotherapy. Methods The expression of NF-κB and LRP mRNA and protein were examined by real time RT-PCR and immunohistochemical assay in 40 specimens of cervicis carcinoma tissue before and after neoadjuvant chemotherapy.Results The expression of NF-κB、LRP mRNA and protein after neoadjuvant chemotherapy were significantly higher than that of before neoadjuvant chemotherapy( P < 0. 01 all), NF-κB was positive correlation with LRP ( r =0. 389, 0. 379, P <0. 05 ) before and after neoadjuvant chemotherapy respective. Conclusions The expression of NF-κB and LRP in cervicis carcinoma tissue may play an important role in the origin development and multi-drug resistance of cervicis carcinoma. Chemotherapy drugs may be induce NF-κB activated and multi-drug resistance.NF-κB activated is correlation with multi-drug resistance of LRP mediated.
