2型糖尿病患者 Klotho 基因 G-395 A 多态性及其对合并下肢动脉粥样硬化的影响

         

摘要

目的:观察2型糖尿病( T2DM)患者Klotho基因G-395A多态性,探讨其对合并下肢动脉粥样硬化的影响。方法采用实时荧光定量聚合酶链式反应( FQ-PCR)技术,分别对205例健康体检者(汉族85例、彝族120例,对照组)、140例T2DM患者(汉族79例、彝族61例,T2DM组)及224例T2DM合并下肢动脉粥样硬化患者(汉族157例、彝族67例,动脉硬化组)的Klotho基因G-395A多态性进行检测,比较各组汉族、彝族基因型频率、等位基因频率,分析T2DM合并下肢动脉粥样硬化的危险因素。结果动脉硬化组汉族Klotho基因G-395A位点GA+AA基因型频率高于彝族(P<0.05),其余各组各等位基因及基因型频率比较差异均无统计学意义(P均>0.05)。汉族T2DM患者发生下肢动脉粥样硬化的影响因素为SBP、TG、LHL、空腹血糖(FPG)、Klotho基因G-395A位点GA+AA基因型(P均<0.05),彝族为TG、LHL、FPG、血肌酐(P均<0.05)。结论 T2DM合并下肢动脉粥样硬化患者存在Klotho基因G-395A多态性,Klotho基因G-395A位点GA+AA基因型可能是云南省汉族部分T2DM患者发生下肢动脉粥样硬化的危险因素。%Objective To observe the G-395A polymorphisms of Klotho gene in type 2 diabetes mellitus ( T2DM) pa-tients, and to investigate its influence on combined lower limb atherosclerosis.Methods The G-395A polymorphisms of Klotho gene from 205 cases of normal controls (85 cases of Han nationality, 120 cases of Yi nationality, control group), 140 cases of T2DM patients (79 cases of Han nationality, 61 cases of Yi nationality, T2DM group) and 224 cases of patients with lower limb atherosclerosis combined with T2DM (157 cases of Han nationality, 67 cases of Yi nationality, atherosclerosis group) were detected by real-time fluorescence quantification PCR ( FQ-PCR) .The genotype frequency and allele frequency were compared between each group, and the risk factors of T2DM combined with lower limb atherosclerosis were analyzed. Results The genotype frequency of Klotho gene G-395A site G/A+A/A in the atherosclerosis group of Han nationality was higher than that of Yi nationality (P<0.05).There was no significant difference in genotype and allele frequency among the rest groups (all P>0.05).The influence factors of T2DM patients with lower extremity atherosclerosis in Han nationality were systolic blood pressure, TG, LHL, fasting blood-glucose ( FPG) and genotype of Klotho gene G-395A site G/A+A/A (all P<0.05).The influence factors of T2DM patients with lower extremity atherosclerosis in Yi nationality were TG, LHL, FPG and serum creatinine (all P<0.05).Conclusions There is gene polymorphism at G-395A of Klotho gene in T2DM patients with lower limb atherosclerosis.The Klotho gene G-395A site G/A+A/A genotype may be a genetic risk factor for the occurrence of lower limb atherosclerosis of T2DM patients in Han nationality.
