急性白血病治疗前后外周血单个核细胞T细胞免疫球蛋白黏蛋白3的表达水平及意义

摘要

目的 探讨急性白血病治疗前后外周血单个核细胞T细胞免疫球蛋白黏蛋白3(Tim-3)的表达水平及意义.方法 选取2015年2月至2017年3月我院收治的25例急性白血病患者(研究组)及同期我院体检的25例健康志愿者及其供体的骨髓(健康组),将单个核细胞分离后提取DNA,受试患者接受标准方案化疗治疗,采用实时荧光定量聚合酶链反应检测单个核细胞上Tim-3 mRNA表达量.结果 研究组治疗前外周血T细胞Tim-3比率及Tim-3 mRNA相对表达量明显高于健康组及治疗后(P＜0.05);而研究组治疗后外周血T细胞Tim-3比率及Tim-3 mRNA相对表达量与健康组差异无统计学意义(P＞0.05);急性白血病患者临床疗效与Tim-3基因表达量呈明显负相关(P＜0.05).结论 Tim-3基因在急性白血病初治患者中存在明显高表达现象,其表达水平高低在急性白血病患者治疗效果的评估中有一定积极作用.%Objective To investigate the expression level and significance of T cell immunoglobulin mucin 3 (Tim-3) in peripheral blood mononuclear ceils before and after treatment in patients with acute leukemia.Methods Twenty-five patients with acute leukemia treated in our hospital from February 2015 to March 2017 were sdected as study group,and 25 healthy volunteers having physical examination or bone marrow of donors during the same period in our hospital were denoted as healthy group.After isolation of mononuclear cells,RNA was extracted and the patients were treated with standard chemotherapy.The expression level of Tim-3 mRNA in monocytes was detected by real-time qPCR.Results The Tim-3 ratio in peripheral blood T cells and the relative expression level of Tim-3 mRNA in the study group before treatment were significantly higher than those in the healthy group and in the study group after treatment group (P ＜ 0.05).The Tim-3 ratio in peripheral blood T cells and the relative expression of Tim-3 mRNA in the study group after treatment were not significantly different from those in the healthy group (P ＞ 0.05).There was a negative correlation between clinical curative effect and expression levels of Tim-3 mRNA.Conclusion Tim-3 gene is highly expressed in patients with acute leukemia.Its expression level plays a role in evaluation of the therapeutic effect of the patients.
