目的:建立西伯利亚接骨木中白桦脂酸的含量测定方法,为完善西伯利亚接骨木的质量标准提供理论依据。方法采用HPLC法。色谱柱:Agilent ® C18(250 mm ×4.6 mm ,5μm);流动相:乙腈‐1 mL · L -1磷酸水溶液(90∶10);柱温:25℃;流速:1.0 mL · min-1;检测波长:210 nm。结果白桦脂酸质量浓度在0.02~0.32 mg · mL -1范围内线性关系良好(r=0.9999),平均回收率为99.6%,RSD值为1.34%(n=9),白桦脂酸平均含量为0.31 mg · g -1。结论该方法简便、准确、可靠,可作为西伯利亚接骨木药材白桦脂酸的含量测定方法。%Objective To consummate the quality standard and to provide theory evidence of Sambucus sibirica Nakai ,by constituting the method of assaying of betulinic acid in Sambucus sibirica Nakai .Methods The content of betulinic acid in Sambucus sibirica Nakai was detected by HPLC .The samples were separated on an Agilent ® C18 column (250 mm × 4 .6 mm ,5 μm) .The mobile phase was acetonitrile‐1 mL·L -1 phosphoric acid (90∶10) .The column temperature was 25 ℃ .The flow rate was 1 .0 mL·min-1 and detected at 210 nm .Results Betulinic acid was linear in the range of 0 .02‐0 .32 mg · mL -1 with a regression coefficient of 0.999 9 and the average recovery was 99 .6% with RSD 1 .34% (n=9) .The average content of betulinic acid was 0 .31 mg · g -1 . Conclusion The determination method was simple ,accurate and reliable ,and could be used as the determination method for betulin‐ic acid in Sambucus sibirica Nakai .
