聚酰胺树脂纯化昆仑雪菊总黄酮的工艺研究

         

摘要

目的:优选聚酰胺树脂分离纯化昆仑雪菊总黄酮的工艺条件。方法采用紫外分光光度法,以总黄酮含量为考察指标,研究在静态和动态条件下,聚酰胺树脂吸附和洗脱总黄酮的行为,筛选出聚酰胺树脂精制雪菊总黄酮的最优工艺参数。结果雪菊总黄酮最佳纯化工艺条件为:60~100目聚酰胺对雪菊总黄酮吸附与解吸较好;清洗液最佳 pH 值为5;乙醇用量为11倍柱体积;乙醇浓度为70%,在该纯化条件下,总黄酮纯度由粗提物22.56%提高到68.04%。结论聚酰胺树脂对雪菊总黄酮的纯化效果好,成本低,可工业化推广。%Objective To optimize purification technique for total flavonoids from Kunlun Chrysanthemum by polyamide resin .Methods With the content of total flavonoids as index ,by using UV spectrophotome‐try ,adsorption and elution pattern of polyamide resin for total flavonoids was investigated under static and dynamic conditions ,and selected optimum technique parameters of purification technology for total fla ‐vonoids by polyamide resin .Results The best parameters for separating the total flavonoids of Kunlun Chrysanthemum :Adsorption and desorption abilities of 60 ~ 100 mech polyamide for total flavonoids were the best of the three polyamides ;Optimum pH value of the cleaning liquid 5 ;the amount of ethanol is 11 column volumes ;ethanol concentration of 70% .Under the purification condition ,the content of total fla‐vonoids could reach 68 .04% from 22 .56% .Conclusion Because of the good effect and low cost ,Polyamide resin for purification of total flavonoids Kunlun Chrysanthemum can be widely used in practice .
