哮喘患儿血浆 IL-4、IFN-γ、SIgE 表达及其与肺功能的相关性研究

         

摘要

目的:研究哮喘患儿血浆白细胞介素-4(IL-4)、γ-干扰素(IFN-γ)、特异性 IgE(SIgE)在儿童哮喘发病中的作用及其与肺功能的相关性。方法采用生物素双抗体夹心酶联免疫吸附法(ELISA)检测90例哮喘急性发作期患儿(哮喘组)及90例年龄、性别相当的健康儿童(对照组)血浆 IL-4、IFN-γ、SIgE 的表达水平,检测哮喘组40例患儿的肺功能1秒钟用力呼气容积(FEV1)、1秒率(FEV1/FVC)、最大呼气流量每日变异率(PEF),并进行相关性分析。结果哮喘组患儿血浆 IL-4水平明显高于对照组,血浆 IFN-γ水平低于对照组,血浆 SIgE 水平高于对照组,差异均有统计学意义(P <0.05)。IL-4、IFN-γ、SIgE 与 FEV1无相关性(P >0.05),IL-4、IFN-γ、SIgE 与PEF 无相关性(P >0.05),IL-4、IFN-γ与 FEV1/FVC 无相关性(P >0.05),SIgE 与 FEV1/FVC 呈正相关(P <0.05)。结论哮喘患儿血浆 IL-4表达增高,血浆 IFN-γ表达降低。SIgE 可为支气管哮喘是否处于急性发作期提供参考。IL-4表达增高及 IFN-γ表达降低共同导致了哮喘炎性过程的发生及发展。%Objective To explore the role of IL-4,IFN-γ and SIgE in plasma of children with asthma and their relationship with the lung functions.Methods The expression levels of IL-4,IFN-γ and SIgE in plasma of 90 children with asthma in attack were detected by ELISA,with the IL-4,IFN-γ and SIgE in plasma of 90 healthy children as the control group.Moreover,lung functions (FEV1,FEV1/FVC and PEF)of 40 children with asthma in attack were measured and correlation analysis of them.Both groups had the same age and sex.Results The results showed that the contents of IL-4 and SIgE in plasma of children with asthma in attack are significant higher than those in serum of healthy children,while the content of IFN-γ in plasma of children with asthma in attack is significant lower than that in serum of healthy children (P <0.05).There were no relationships of IL-4,IFN-γand SIgE with the FEV1 and PEF (P >0.05).FEV1/FVC had significant positive correlation with SIgE (P <0.05),but had no correlation with IL-4 and IFN-γ(P >0.05).Conclusion The expression level of IL-4 in plasma of children with asth-ma in attack is increased.The expression level of IFN-γin plasma of children with asthma in attack is de-creased.SIgE is the theoretical basis for the acute attack of bronchial asthma.The increased expression lev-el of IL-4 and decreased expression level of IFN-γ together result in the occurrence and development of asthma in the inflammatory process.
