Notch 信号通路在肝癌细胞迁移过程中的作用及机制

         

摘要

Objective:To explore the mechanism of Notch signaling pathway in the migration of hepatocellular car-cinoma cells. Methods:Cultured human hepatoma SMMC - 7721,MHCC97H and normal liver HL - 7702 cells in vitro,using transwell assay to test the ability of cell migration. Using Western blot to detect the protein expression of Notch1,Snail,E - cadherin and COX - 2. Compared the cell migration ability before and after blocking Notch signa-ling pathway with 5μmol/ L DAPT(γ secretase inhibitors)and blocking COX - 2 with 50μmol/ L NS - 398. Results:Notch1,Snail,E - cadherin and COX - 2 were involved in the migration process of HCC cells. The effect of DAPT could up - regulate the expression of E - cadherin and down - regulate the expression of Snail and COX - 2. Conclu-sion:The role of Notch signaling pathway in HCC cells migration is very important,and the mechanism maybe related to the levels of Snail,E - cadherin and COX - 2.%目的:探讨 Notch 信号通路在肝癌细胞迁移过程中的作用及相关机制。方法:体外培养肝癌细胞系SMMC -7721、MHCC97H 和正常非肿瘤肝细胞系 HL -7702,利用 Transwell 小室检测细胞的迁移能力,Western blot 检测 Notch1、Snail、E - cadherin、COX -2(环氧化酶-2)蛋白的表达。分别采用5μmol/ L DAPT(3,5-二氟苯乙酰- L -丙氨酰- S 苯基甘氨酸 t -丁酯,γ-分泌酶抑制剂)阻断 Notch 信号通路和50μmol/ L NS -398阻断 COX -2,比较对细胞迁移能力的影响以及对肝癌细胞中 Snail、E - cadherin 表达的影响。结果:Notch1、Snail、E - cadherin、COX -2参与了肝癌细胞迁移过程,DAPT 通过阻断 Notch 显著上调 E - cadherin,影响细胞的迁移(P <0.05),下调 Snail 和 COX -2,NS -398通过阻断 COX -2影响细胞的迁移(P <0.05)。结论:Notch 信号通路在肝癌细胞迁移过程中起着非常重要的作用,其机制初步认为与 Snail、E - cadherin、COX -2的表达有关。
